Overview
Lamotrigine + bupropion is a combination therapy consisting of lamotrigine, a phenyltriazine anticonvulsant, and bupropion, an aminoketone antidepressant. Lamotrigine acts primarily by inhibiting voltage-gated sodium channels, which stabilizes neuronal membranes and inhibits the release of excitatory neurotransmitters, particularly glutamate. Bupropion functions as a norepinephrine-dopamine reuptake inhibitor (NDRI) and a non-competitive antagonist of nicotinic acetylcholine receptors. This combination is frequently utilized in psychiatric practice for the management of bipolar depression and treatment-resistant major depressive disorder. The mood-stabilizing properties of lamotrigine complement the antidepressant effects of bupropion, potentially providing a synergistic therapeutic benefit while mitigating the risk of antidepressant-induced manic switching in patients with bipolar disorder.
