Overview

**Lanreotide + pegvisomant** is a combination therapy used in the treatment of acromegaly, particularly for patients who are not adequately controlled with monotherapy. Lanreotide is a long-acting somatostatin analog that inhibits growth hormone (GH) secretion from pituitary adenomas by binding to somatostatin receptors, reducing GH and subsequently IGF-1 production. Pegvisomant is a pegylated recombinant growth hormone receptor antagonist; it blocks the action of GH at the receptor level in peripheral tissues, directly reducing the synthesis of IGF-1 in the liver. The combination is generally well tolerated and can achieve normalization of IGF-1 levels in most patients, often requiring lower doses of pegvisomant compared to monotherapy. This therapy is not a fixed drug combination but rather a clinical co-administration strategy.[1][2][3]
