LAP (mut) IL-2 T pep is a novel recombinant fusion protein designed to deliver interleukin-2 (IL-2) selectively to the tumor microenvironment. Developed by Stealthyx Therapeutics, the molecule consists of IL-2 sequestered within a protective shell formed by the Latency Associated Peptide (LAP) of the TGF-β precursor. The two components are joined by a linker containing a matrix metalloproteinase (MMP) cleavage site. This configuration is intended to mask the biological activity of IL-2 systemically, thereby extending its half-life and reducing the severe vascular leak syndrome and other toxicities associated with high-dose aldesleukin. Upon reaching the tumor site, where MMP levels are typically elevated, the linker is cleaved, releasing active IL-2 to stimulate tumor-infiltrating lymphocytes. Preclinical studies in syngeneic mouse models of melanoma and renal cell carcinoma have demonstrated significant anti-tumor activity and improved tolerability compared to standard recombinant IL-2.
