Lapatinib + trametinib is an investigational combination therapy consisting of two small molecule kinase inhibitors, lapatinib and trametinib. Lapatinib is a reversible inhibitor of the intracellular tyrosine kinase domains of both epidermal growth factor receptor (EGFR/HER1/ERBB1) and human epidermal growth factor receptor type 2 (HER2/ERBB2), blocking downstream signaling pathways involved in tumor cell proliferation and survival[1][4]. Trametinib is a highly selective, allosteric inhibitor of MEK1 and MEK2, key kinases in the MAPK/ERK pathway that mediates cellular proliferation[6]. The rationale for combining these agents is to achieve dual inhibition of EGFR/HER2-driven signaling as well as the MAPK pathway, which may be particularly relevant for tumors with KRAS mutations or resistance to single-agent therapies. This combination has been studied in phase I clinical trials for advanced solid tumors such as colorectal cancer, non-small cell lung cancer (NSCLC), and pancreatic cancer with specific molecular profiles[3][5].
