**Laromustine + hydroxyurea** is an experimental combination therapy of two small molecule cytotoxic agents investigated in cancer treatment. **Laromustine** (also known as VNP40101M, Onrigin, cloretazine) is an alkylating agent of the nitrosourea class, which induces DNA cross-linking and inhibits DNA synthesis and function, ultimately resulting in cell death. **Hydroxyurea** is an antimetabolite that inhibits ribonucleotide reductase, reducing deoxyribonucleotide pools required for DNA synthesis, exerting synergistic cytotoxic effects, especially against rapidly dividing cells. This combination has been primarily evaluated preclinically and in early-stage clinical investigations for potential anti-tumor synergy, notably in hematologic malignancies and solid tumors such as gliomas. Each drug operates by distinct but complementary mechanisms resulting in augmented cytotoxicity.
