Overview

LASSARAB + aPHAD-SEat is an investigational combination vaccine candidate designed to protect against both Lassa fever and rabies. The product consists of an inactivated, recombinant rabies virus vector expressing the codon-optimized glycoprotein complex (GPC) of the Lassa virus (LASV), as well as the rabies glycoprotein (RABV G). The vaccine is formulated with 3D-(6-Acyl) PHAD-SE, an oil-in-water emulsion adjuvant that acts as a toll-like receptor 4 (TLR4) agonist to enhance immune responses. Preclinical studies have shown that this combination elicits robust humoral immunity against both LASV and RABV, providing protection in animal models. The mechanism of protection involves induction of non-neutralizing antibodies targeting LASV GPC, which mediate antibody-dependent cellular functions critical for defense against Lassa fever[1][2][6]. Developed by Thomas Jefferson University, this dual-purpose vaccine aims to address major unmet needs in West Africa where both diseases are endemic[7].
