Overview

LAVA-051 + interleukin-2 is an experimental combination therapy designed for the treatment of hematological malignancies. LAVA-051 is a humanized bispecific single-domain antibody (Gammabody) that targets both the Vγ9Vδ2 T cell receptor and CD1d on tumor cells, engaging both Vγ9Vδ2 T cells and type 1 NKT cells to induce potent antitumor effects while aiming to reduce off-tumor toxicity. Interleukin-2 (IL-2) is a recombinant cytokine used clinically to promote T-cell proliferation and activation. The combination is rationalized by the ability of IL-2 to expand and activate Vγ9Vδ2 T cells, enhancing the activity of LAVA-051. The individual components are being studied in relapsed/refractory chronic lymphocytic leukemia (CLL), multiple myeloma (MM), and acute myeloid leukemia (AML), though the combination of low-dose IL-2 with LAVA-051 is specifically noted as a planned or preclinical cohort rather than an extensively trialed arm[3]. LAVA Therapeutics N.V. is the primary developer of LAVA-051.
