LBL-024 + etoposide + cisplatin is a combination therapy under development for advanced extrapulmonary neuroendocrine carcinoma (EP-NEC). LBL-024 is a first-in-class bispecific antibody that targets Programmed Death Ligand-1 (PD-L1) and the co-stimulatory receptor 4-1BB. By inhibiting PD-L1 and co-stimulating 4-1BB, LBL-024 both blocks immunosuppressive signaling and enhances T-cell activation in the tumor microenvironment, resulting in a potent anti-tumor immune response. When combined with etoposide (a topoisomerase II inhibitor) and cisplatin (a DNA crosslinking agent/platinum-based chemotherapy), the regimen aims to deliver synergistic cytotoxicity and immune-mediated tumor destruction. Recent phase Ib/II clinical trial data show robust antitumor activity and tolerability when this combination is used as first-line therapy for advanced EP-NEC. LBL-024 is developed by Leads Biolabs. Etoposide and cisplatin are established chemotherapeutic agents with multiple manufacturers.
