LD + GC012F

LD + GC012F is a therapeutic regimen consisting of lymphodepletion (LD) preconditioning followed by the infusion of GC012F (AZD0120), an autologous dual-targeting chimeric antigen receptor T-cell (CAR-T) therapy. GC012F simultaneously targets CD19 and B-cell maturation antigen (BCMA) to achieve comprehensive depletion of B cells and plasma cells. Developed by Gracell Biotechnologies (acquired by AstraZeneca) using the proprietary FasTCAR-T platform, the therapy features a rapid next-day manufacturing process. The regimen is being investigated for the treatment of refractory autoimmune diseases, including systemic lupus erythematosus (SLE) and difficult-to-treat (D2T) rheumatoid arthritis, as well as hematologic malignancies like multiple myeloma. The lymphodepletion component typically involves chemotherapy agents such as cyclophosphamide and fludarabine to create a favorable environment for CAR-T cell expansion and persistence.
