Overview

Lemzoparlimab + azacitidine is an investigational combination therapy for hematologic malignancies, particularly higher-risk myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML). Lemzoparlimab is a novel humanized monoclonal antibody targeting CD47, a cell surface protein that delivers a "don't eat me" signal to macrophages. By blocking CD47, lemzoparlimab enables macrophage-mediated phagocytosis of leukemic cells. Azacitidine is a well-established hypomethylating agent used in the treatment of MDS and AML. The combination has demonstrated strong anti-leukemic activity in preclinical studies and early-phase clinical trials, with encouraging response rates and manageable safety profiles. Lemzoparlimab’s design aims to minimize binding to normal red blood cells, potentially reducing the risk of severe anemia compared to other anti-CD47 antibodies[1][2][5].
