**Lenalidomide + dacarbazine** is an investigational combination therapy composed of two drugs: lenalidomide, a thalidomide derivative with immunomodulatory, anti-angiogenic, and antineoplastic activity, and dacarbazine, an alkylating agent that induces cancer cell death by causing DNA breaks. Lenalidomide modulates the immune system by enhancing anti-inflammatory cell populations and natural killer cell cytotoxicity, while inhibiting pro-inflammatory cytokines and altering transcription factors through degradation of IKZF1, IKZF3, and CK1α. Dacarbazine is a cytotoxic agent that works by alkylating DNA, leading to apoptosis in rapidly dividing cancer cells. This combination has primarily been investigated in phase I/II trials for metastatic malignant melanoma and other cancers, aiming to improve antitumor efficacy over single-agent treatments.
