RAD is a combination chemotherapy regimen consisting of lenalidomide, adriamycin (doxorubicin), and dexamethasone. Developed by Wuerzburg University Hospital in collaboration with the Deutsche Studiengruppe Multiples Myelom (DSMM), it is primarily utilized as an induction therapy for patients with newly diagnosed multiple myeloma, often as a precursor to risk-adapted stem cell transplantation. Lenalidomide serves as an immunomodulatory agent that targets the cereblon E3 ubiquitin ligase complex, doxorubicin is a cytotoxic anthracycline that inhibits topoisomerase II and intercalates DNA, and dexamethasone is a potent corticosteroid that provides anti-inflammatory effects and enhances the pro-apoptotic activity of the other agents. The regimen is typically administered in 28-day cycles and has been evaluated in Phase II and Phase III clinical trials.
