Overview

Lenalidomide + Zn-DDC is an investigational combination therapy consisting of the immunomodulatory drug lenalidomide and the zinc-chelating compound zinc diethyldithiocarbamate (Zn-DDC). This combination is being evaluated for its potential to target the Hypoxia-NFκB-Cancer Stem Cell (CSC) pathway in haematological malignancies, specifically multiple myeloma. Lenalidomide acts by binding to the cereblon E3 ubiquitin ligase complex, leading to the degradation of transcription factors Ikaros (IKZF1) and Aiolos (IKZF3), while Zn-DDC (also known as Imuthiol) has demonstrated toxicity toward cancer stem cells. The combination is currently being studied in translational bench studies using patient bone marrow samples to improve treatment outcomes in relapsed or refractory multiple myeloma.
