Overview

Lenvatinib + envafolimab is a combination therapy under investigation for the treatment of various advanced cancers, including unresectable hepatocellular carcinoma (HCC) and endometrial cancer. Lenvatinib is an oral small molecule receptor tyrosine kinase inhibitor that targets multiple kinases involved in tumor angiogenesis and growth, such as VEGFR1–3, FGFR1–4, PDGFRα, KIT, and RET. Envafolimab is a monoclonal antibody immune checkpoint inhibitor targeting PD-L1; it is administered subcutaneously. The combination leverages antiangiogenic effects from lenvatinib with immune activation from envafolimab to enhance antitumor activity. Clinical studies have shown promising efficacy and manageable safety profiles for this regimen in patients with advanced HCC and endometrial cancer who have progressed after prior therapies[2][5][7].
