Overview

Lenvatinib + gemcitabine + oxaliplatin is a combination regimen consisting of three drugs: lenvatinib, a multi-targeted tyrosine kinase inhibitor with antiangiogenic and antiproliferative properties; gemcitabine, a nucleoside analog that inhibits DNA synthesis; and oxaliplatin, a platinum-based chemotherapeutic agent that causes DNA crosslinking and apoptosis. This combination has been investigated primarily for the treatment of advanced or unresectable fibrolamellar carcinoma (FLC) and intrahepatic cholangiocarcinoma (ICC), particularly in patients who are not candidates for surgery or have relapsed/progressed after prior therapies. The rationale for this regimen is to combine the cytotoxic effects of chemotherapy (gemcitabine/oxaliplatin) with the targeted inhibition of tumor angiogenesis and growth signaling pathways by lenvatinib[1][2][3].
