Lenvatinib + pembrolizumab + pemetrexed + carboplatin is an investigational four-drug combination regimen developed by Eisai and Merck & Co. for the first-line treatment of metastatic nonsquamous non-small cell lung cancer (NSCLC). The regimen combines lenvatinib (a multi-kinase inhibitor targeting VEGFR and FGFR), pembrolizumab (an anti-PD-1 monoclonal antibody), and a chemotherapy backbone of pemetrexed and carboplatin. The combination was designed to enhance anti-tumor activity by simultaneously targeting angiogenesis, immune evasion, and cellular proliferation. However, the Phase 3 LEAP-006 trial (NCT03829319) demonstrated that the addition of lenvatinib to the pembrolizumab plus chemotherapy standard-of-care did not provide a survival benefit, leading to the termination of this specific combination's development for this indication.
