Overview

This is a combination therapy consisting of three agents: **Lenvatinib** is a small molecule multi-targeted tyrosine kinase inhibitor that blocks the activity of several receptor tyrosine kinases involved in tumor angiogenesis and proliferation, including vascular endothelial growth factor receptors (VEGFR1, VEGFR2, VEGFR3), fibroblast growth factor receptors (FGFR1–4), platelet-derived growth factor receptor alpha (PDGFRα), KIT, and RET. It is approved for use in various cancers such as thyroid cancer, renal cell carcinoma, and hepatocellular carcinoma[1][2][5]. **Sindilimab** appears to be a misspelling or variant spelling of "sintilimab," which is a humanized monoclonal antibody targeting PD-1; however, no direct evidence for "sindilimab" as an approved or investigational agent was found. **Carrelizumab**, more commonly spelled "camrelizumab," is a humanized anti-PD-1 monoclonal antibody that blocks the interaction between PD-1 on T cells and its ligands PD-L1/PD-L2 on tumor cells. This releases the inhibition on T cells and enhances immune-mediated tumor cell killing[6][9]. Camrelizumab has been developed primarily by Jiangsu Hengrui Pharmaceuticals and is approved in China for several cancers including hepatocellular carcinoma[9]. The combination of these agents would theoretically provide both antiangiogenic effects via lenvatinib’s inhibition of multiple pro-tumorigenic kinases and enhanced antitumor immunity through dual checkpoint blockade with two anti-PD-1 antibodies.
