Overview

Lenvatinib + sintilimab + SIRT is a combination therapy consisting of lenvatinib, a multi-kinase inhibitor; sintilimab, an anti-PD-1 monoclonal antibody; and selective internal radiation therapy (SIRT), also known as radioembolization. Lenvatinib inhibits multiple receptor tyrosine kinases involved in tumor angiogenesis and proliferation. Sintilimab blocks the PD-1 pathway to enhance anti-tumor immune responses. SIRT delivers targeted internal radiation directly to liver tumors via microspheres loaded with radioactive isotopes, typically yttrium-90. This triple regimen is under investigation for advanced hepatocellular carcinoma (HCC) and other liver cancers, aiming to leverage synergistic effects of targeted therapy, immunotherapy, and locoregional radiotherapy.
