Lenvatinib + VIC-1911 is an investigational combination therapy for advanced hepatocellular carcinoma (HCC), particularly in patients who are unresponsive or resistant to lenvatinib monotherapy. Lenvatinib is a small molecule multikinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR1–3), fibroblast growth factor receptors (FGFR1–4), platelet-derived growth factor receptor alpha (PDGFRα), RET proto-oncogene, and KIT proto-oncogene. It inhibits tumor angiogenesis and proliferation. VIC-1911 (also known as TAS-119 or TAS-2104) is a highly selective oral small molecule inhibitor of aurora kinase A (AURKA), which plays a critical role in mitotic spindle formation and cell division; its inhibition leads to cell cycle arrest and apoptosis in cancer cells. The combination aims to overcome resistance mechanisms by targeting both angiogenesis pathways and mitotic regulation[2][3][10].
