Overview
Lerapolturev + lomustine is an investigational drug combination being evaluated in clinical trials for the treatment of recurrent glioblastoma. Lerapolturev (also known as PVSRIPO) is a genetically modified oncolytic poliovirus designed to induce antitumor immune response by infecting and lysing tumor cells and stimulating local immune activation. Lomustine is a small molecule chemotherapy agent from the nitrosourea class, which acts as an alkylating agent that cross-links and alkylates DNA, leading to cancer cell death. Lerapolturev is administered intratumorally via convection enhanced delivery, followed by subcutaneous injection near cervical lymph nodes, while lomustine is given orally. The combination is being studied for efficacy and safety in adults with recurrent glioblastoma after maximal safe resection, with each drug arm administered separately in the study protocol[1][3][5].
