Overview
lesinurad + xanthine oxidase inhibitor refers to the combination therapy developed and formerly commercialized by Ironwood Pharmaceuticals for the treatment of hyperuricemia associated with gout. This therapeutic approach combines lesinurad, a selective uric acid reabsorption inhibitor (SURI) that targets the URAT1 and OAT4 transporters in the kidney to increase uric acid excretion, with a xanthine oxidase inhibitor (XOI) such as allopurinol or febuxostat, which reduces uric acid production. The combination was designed for patients who had not achieved target serum uric acid levels with XOI monotherapy. Ironwood marketed lesinurad as Zurampic and a fixed-dose combination with allopurinol as Duzallo before discontinuing its gout portfolio and returning the rights to AstraZeneca in 2019.
