Letetresgene autoleucel (lete-cel; GSK3377794) is a genetically modified autologous T-cell therapy engineered to express an enhanced T-cell receptor (TCR) specific for the cancer-testis antigens NY-ESO-1 and LAGE-1a. It is designed to target and eliminate tumor cells expressing these antigens in patients who are HLA-A*02:01, *02:05, or *02:06 positive. Pembrolizumab (Keytruda) is a monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1), thereby enhancing immune responses against tumors. The combination of letetresgene autoleucel with pembrolizumab has been investigated in clinical trials for relapsed/refractory multiple myeloma and advanced non-small cell lung cancer, as well as other solid tumors such as myxoid/round cell liposarcoma. Letetresgene autoleucel has demonstrated manageable safety and transient antitumor activity, while pembrolizumab provides additional immune checkpoint inhibition[1][5][6][8].
