letrozole + berberine

Letrozole + berberine is a combination of two small molecule agents, letrozole and berberine, investigated for synergistic therapeutic effects in several indications. Letrozole is a nonsteroidal aromatase inhibitor that reduces estrogen production by inhibiting the aromatase enzyme, primarily used in hormone receptor-positive breast cancer and ovulation induction. Berberine is a plant-derived isoquinoline alkaloid with multiple pharmacological activities, including antidiabetic, anti-inflammatory, and anticancer effects; it modulates various molecular targets such as AMP-activated protein kinase (AMPK) and has been used to improve insulin resistance. The combination has been studied for enhanced cytotoxicity against breast cancer cells via nanoparticle delivery systems[1][2], as well as for improving ovulation and live birth rates in women with polycystic ovary syndrome (PCOS)-related infertility[3][5]. The rationale behind the combination includes potential synergistic anticancer activity and improved metabolic/hormonal profiles.
