Overview

Letrozole + goserelin is a combination of two pharmaceutical agents used primarily in the treatment of hormone receptor-positive advanced or metastatic breast cancer in premenopausal women. Letrozole is a nonsteroidal aromatase inhibitor that suppresses estrogen synthesis by inhibiting the aromatase enzyme, thereby reducing circulating estrogen levels. Goserelin is a luteinizing hormone-releasing hormone (LHRH) agonist that acts on the pituitary gland to downregulate gonadotropin secretion, leading to ovarian suppression and decreased production of endogenous estrogens. This combination effectively induces chemical menopause and further reduces estrogen stimulation of breast cancer cells, making it an effective endocrine therapy for premenopausal women with advanced disease[1][2][3]. The regimen has demonstrated efficacy and tolerability as first- or second-line hormonal therapy in clinical studies.
