Letrozole + palbociclib is a fixed combination therapy used as initial endocrine-based treatment for hormone receptor–positive, human epidermal growth factor receptor 2 (HER2)-negative advanced or metastatic breast cancer in adults. Letrozole is an aromatase inhibitor that reduces estrogen production, thereby limiting the growth stimulus for estrogen-dependent breast cancer cells. Palbociclib is a selective cyclin-dependent kinase 4/6 (CDK4/6) inhibitor that blocks cell cycle progression from G1 to S phase by preventing phosphorylation of the retinoblastoma protein, leading to inhibition of tumor cell proliferation.
