Overview
A combination therapy consisting of letrozole, an aromatase inhibitor, and patritumab deruxtecan, a HER3-directed antibody-drug conjugate. Patritumab deruxtecan consists of a fully human monoclonal antibody against HER3 attached to a topoisomerase I inhibitor payload (DXd, an exatecan derivative) via a tetrapeptide-based cleavable linker. The combination is being investigated for the treatment of hormone receptor-positive (HR+), HER2-negative breast cancer, where it has shown comparable efficacy to multiagent chemotherapy with fewer side effects.
