Letrozole + progesterone is a combination of two pharmaceutical agents used primarily in reproductive medicine and gynecology. Letrozole is a non-steroidal aromatase inhibitor that reduces estrogen synthesis by inhibiting the enzyme aromatase, thereby lowering circulating estrogen levels. This mechanism is particularly useful for ovulation induction in women with infertility, such as those with polycystic ovarian syndrome (PCOS), and for hormone-sensitive breast cancer treatment[1][4][6]. Progesterone is a naturally occurring steroid hormone essential for regulating the menstrual cycle and maintaining pregnancy; it acts on progesterone receptors to prepare the endometrium for implantation and support early gestation[8]. The combination of letrozole with progestins (such as dydrogesterone or other forms of progesterone) has been studied in conditions like endometriosis, where it can modulate sex hormone levels, reduce inflammatory markers, and improve clinical outcomes[3]. In assisted reproductive technology (ART), this combination may be used to induce ovulation while providing luteal phase support.
