Letrozole + temsirolimus

Letrozole + temsirolimus is a combination of two pharmaceutical agents used in clinical research for the treatment of advanced, hormone receptor–positive breast cancer. Letrozole is a nonsteroidal aromatase inhibitor that reduces estrogen production by inhibiting the aromatase enzyme, thereby decreasing tumor growth in estrogen-dependent cancers[2]. Temsirolimus is an mTOR (mammalian target of rapamycin) inhibitor that binds to FKBP-12 and inhibits mTOR activity, leading to cell cycle arrest and reduced tumor proliferation[4][6][8]. The combination has been studied as first-line therapy in postmenopausal women with advanced breast cancer; however, phase III trials did not show significant improvement in progression-free survival compared to letrozole alone[3][5].
