Overview

Letrozole + vinorelbine is a combination therapy consisting of letrozole, an aromatase inhibitor, and vinorelbine, a semi-synthetic vinca alkaloid chemotherapy agent. Letrozole works by inhibiting the aromatase enzyme, thereby reducing estrogen production and limiting the growth of hormone receptor-positive (HR+) breast cancer cells. Vinorelbine disrupts microtubule formation during cell division by binding to tubulin, leading to mitotic arrest and apoptosis in rapidly dividing tumor cells. This combination has been studied primarily in postmenopausal women with HR+/HER2-negative early or advanced breast cancer. Clinical studies have shown that the combination produces a superior anti-proliferative effect compared to either drug alone in this population[1][4]. The regimen is generally well-tolerated with manageable side effects such as arthralgias, asthenia, diarrhea, hot flushes, and nausea[1]. The combination may also enhance immune-related biological processes within tumors[1].
