Overview

**leucine + metformin** is a combination therapy consisting of the **branched-chain amino acid leucine** and the **antidiabetic agent metformin**. Leucine functions primarily by activating the mTORC1 signaling pathway to promote skeletal muscle protein synthesis, improve muscle anabolism, and modulate lipid metabolism. Metformin acts by inhibiting mitochondrial respiratory chain complex I, increasing cellular AMP/ATP ratio and secondarily activating AMP-activated protein kinase (AMPK), thus decreasing hepatic glucose production, improving peripheral insulin sensitivity, and enhancing GLUT4 translocation. The combination has been investigated for **synergistic effects in metabolic optimization, muscle atrophy prevention, cardiometabolic disorders, and obesity**. Metformin and leucine together have demonstrated the ability to attenuate muscle fiber atrophy and reduce cellular senescence markers in preclinical and clinical studies. Both agents target complementary pathways: metformin primarily addresses insulin resistance and gluconeogenesis, while leucine supports muscle protein synthesis and may counteract the catabolic effects of metformin or caloric restriction[1][2]. This combination may be used alone or as part of more complex metabolic therapies such as the triple agent NS-0200 (which includes sildenafil)[4][5].
