Overview
Leucine + sildenafil is a fixed-dose combination of the amino acid L-leucine and the phosphodiesterase type 5 (PDE5) inhibitor sildenafil. This investigational therapy was developed to target metabolic diseases such as obesity by modulating energy metabolism. The mechanism involves allosteric activation of sirtuin 1 (SIRT1) by leucine and inhibition of PDE5A by sildenafil, which together are thought to synergistically activate AMP-activated protein kinase (AMPK), SIRT1, and nitric oxide pathways. These pathways play key roles in regulating energy expenditure and fat metabolism. Clinical studies have evaluated this combination for its effects on body weight reduction in obese individuals[1][3][4].
