Leuprolide + triptorelin is a combination of two synthetic peptide analogs of gonadotropin-releasing hormone (GnRH), both classified as GnRH agonists. Each drug acts by initially stimulating, then profoundly suppressing, the pituitary release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), leading to decreased production of sex hormones such as testosterone and estradiol. Both agents are used individually for conditions like advanced prostate cancer, central precocious puberty, endometriosis, uterine fibroids, and in assisted reproductive technology protocols. The combination itself is not a standard or approved pharmaceutical product but would theoretically provide additive or redundant suppression of the hypothalamic-pituitary-gonadal axis through the same mechanism[1][4][5][6][7]. There is no evidence that this specific combination offers clinical benefit over monotherapy with either agent.
