Overview

Levobupivacaine + mepivacaine is an investigational combination of two amide-type local anesthetics used for regional anesthesia and postoperative pain management. Levobupivacaine is the S(-)-enantiomer of bupivacaine, providing long-acting sensory and motor blockade, while mepivacaine is an intermediate-acting agent. Both drugs function by binding to and inhibiting voltage-gated sodium channels (Nav) on the neuronal membrane, preventing the initiation and conduction of nerve impulses. This combination is primarily studied in the context of nerve blocks (such as femoral, saphenous, or i-pack blocks) for patients undergoing total knee replacement surgery to optimize analgesic efficacy and recovery.
