Lexatumumab + interferon gamma 1b is an experimental combination therapy investigated by the National Cancer Institute (NCI) for the treatment of refractory pediatric solid tumors, including Ewing's sarcoma, osteosarcoma, neuroblastoma, and rhabdomyosarcoma. Lexatumumab (HGS-ETR2) is a fully human monoclonal antibody that acts as an agonist for TRAIL receptor 2 (DR5), inducing apoptosis in tumor cells. However, many pediatric tumors exhibit resistance to TRAIL-mediated apoptosis due to low expression of caspase-8. Interferon gamma 1b, a recombinant cytokine, is added to the regimen to upregulate caspase-8 expression, thereby restoring the sensitivity of the tumor cells to lexatumumab-induced cell death. This synergistic approach was evaluated in a Phase I dose-escalation trial (NCT00428272).
