**LGG@Ga-poly** is an orally administered, preclinical engineered probiotic therapeutic consisting of *Lactobacillus rhamnosus* GG functionalized with a gallium-polyphenol network. It is designed for pancreatic ductal adenocarcinoma by preferentially targeting pancreatic tumors and disrupting iron respiration in tumor-promoting intratumoral Proteobacteria. Depletion of these bacteria and microbiota-derived lipopolysaccharides reduces tumor Toll-like receptor signaling, suppresses PD-L1 and interleukin-1β expression, decreases immunotolerant myeloid populations, enhances cytotoxic T-cell infiltration, inhibits tumor growth, and improves immune-checkpoint-blockade activity in female mouse models.
