Overview

Lidocaine + prilocaine is a fixed-dose combination of two amide-type local anesthetics. Both agents act by blocking voltage-gated sodium channels in neuronal cell membranes, thereby inhibiting the initiation and conduction of nerve impulses. This results in reversible local anesthesia (numbing) at the site of application. The combination is most commonly formulated as a topical cream or patch (e.g., EMLA cream), used for dermal anesthesia prior to minor procedures such as intravenous catheter insertion, blood sampling, superficial surgical interventions, skin grafts, laser hair removal, tattooing preparation, and cleansing or debridement of leg ulcers. It can also be used on genital mucosa for minor procedures or to treat premature ejaculation via topical spray formulations[1][6][9]. Onset and duration depend on dose and application time; effective anesthesia typically requires 1–2 hours under occlusion for intact skin[1][6]. Systemic absorption is generally low but increases with larger doses or compromised skin integrity.
