Overview
Lifileucel + pembrolizumab is a combination therapy under investigation for advanced melanoma and other solid tumors. Lifileucel is an autologous tumor-infiltrating lymphocyte (TIL) cell therapy, where TILs are harvested from a patient's own tumor, expanded ex vivo, and then reinfused to target cancer cells. Pembrolizumab is a monoclonal antibody that blocks the programmed cell death protein 1 (PD-1) receptor on T cells, enhancing immune response against tumors. The combination aims to leverage the direct cytotoxicity of expanded TILs with the immune checkpoint blockade provided by pembrolizumab. This regimen has shown promising efficacy in immunotherapy-naive advanced melanoma patients, with high objective and complete response rates in early-phase trials[1][4][8]. Lifileucel was developed by Iovance Biotherapeutics; pembrolizumab was developed by Merck & Co.
