Overview

Limertinib + radiotherapy is a combination therapeutic regimen being investigated for the treatment of EGFR-mutant non-small cell lung cancer (NSCLC) with brain metastases. Limertinib (ASK120067) is an orally bioavailable, third-generation, irreversible tyrosine kinase inhibitor (TKI) that selectively targets both EGFR-sensitizing mutations and the T790M resistance mutation while sparing wild-type EGFR. Developed by Jiangsu Aosaikang Pharmaceutical and Innovent Biologics, limertinib is designed with high central nervous system (CNS) penetration. Radiotherapy is used in conjunction with the TKI to provide local control of brain lesions, potentially acting synergistically to improve intracranial response rates and progression-free survival. This specific combination is currently being evaluated in a Phase II clinical trial led by The First Affiliated Hospital of Guangzhou Medical University.
