linperlisib + adebrelimab

Linperlisib + adebrelimab is an investigational combination therapy being evaluated for the treatment of advanced biliary tract cancer (BTC). Linperlisib is a highly selective, small-molecule inhibitor of the delta isoform of phosphoinositide 3-kinase (PI3Kδ), which plays a crucial role in B-cell proliferation and survival, as well as the modulation of the tumor immune microenvironment by inhibiting regulatory T cells (Tregs). Adebrelimab is a humanized monoclonal antibody targeting programmed death-ligand 1 (PD-L1), designed to block the interaction between PD-L1 and its receptor PD-1, thereby restoring T-cell-mediated anti-tumor immune responses. The rationale for this combination lies in the potential synergistic effect of PI3Kδ inhibition—which can reduce immunosuppressive cells in the tumor microenvironment—and PD-L1 blockade, which enhances effector T-cell activity. Clinical trials, including those led by the Third Affiliated Hospital of Naval Medical University, are investigating this regimen in patients with refractory or advanced biliary tract malignancies.
