Linperlisib + CPT is a combination therapeutic regimen consisting of linperlisib, a selective small molecule inhibitor of the delta isoform of phosphatidylinositol 3-kinase (PI3Kδ), and the CPT regimen, which includes cyclophosphamide, prednisone, and thalidomide. Linperlisib, developed by Shanghai Yingli Pharmaceutical, targets the PI3Kδ pathway, which is critical for the proliferation and survival of malignant B-cells and T-cells. Cyclophosphamide acts as an alkylating agent that cross-links DNA, while prednisone is a corticosteroid with lympholytic activity. Thalidomide provides immunomodulatory effects by targeting cereblon (CRBN) and inhibiting angiogenesis. This specific combination is being evaluated in a multicenter Phase 2 clinical trial sponsored by The First Affiliated Hospital with Nanjing Medical University for the treatment of various relapsed or refractory non-Hodgkin lymphoma (NHL) subtypes, including follicular lymphoma, peripheral T-cell lymphoma, NK/T-cell lymphoma, chronic lymphocytic leukemia, marginal zone lymphoma, and mantle cell lymphoma.
