Overview

Linperlisib + SHR2554 is an investigational combination therapy comprising two oral small molecule inhibitors for the treatment of relapsed or refractory peripheral T-cell lymphoma (PTCL) and other mature lymphoid neoplasms. Linperlisib is a highly selective phosphoinositide 3-kinase delta (PI3Kδ) inhibitor, while SHR2554 is an enhancer of zeste homolog 2 (EZH2) inhibitor. The combination aims to target both PI3Kδ-mediated signaling pathways involved in tumor cell proliferation and survival as well as epigenetic regulation via EZH2 inhibition, which may enhance antitumor activity in hematologic malignancies. Both agents have demonstrated promising efficacy and safety profiles individually; their combination is currently being evaluated in early-phase clinical trials for PTCL[1][2][3][4].
