Linsitinib + paclitaxel is an investigational **drug combination** of linsitinib, an orally available small molecule dual inhibitor of insulin-like growth factor 1 receptor (IGF-1R) and insulin receptor (IR), and paclitaxel, a well-established microtubule-stabilizing chemotherapy agent. This combination has been investigated in clinical trials for **advanced solid tumors**, especially platinum-resistant/refractory ovarian cancer. Preclinical studies support their combination via inhibition of IGF-1R signaling pathways implicated in chemotherapy resistance. Multiple phase 1 and phase 2 studies evaluated safety and efficacy, but clinical results for ovarian cancer showed no improvement in outcomes compared to paclitaxel alone. Linsitinib has separately shown promising activity in thyroid eye disease, but the combination itself remains under investigation for cancer.
