Overview

Lintuzumab + cytarabine is a combination therapy investigated in older adults with previously untreated acute myeloid leukemia (AML). Lintuzumab (SGN-33, HuM195) is a humanized monoclonal antibody targeting CD33, a cell surface antigen expressed on most AML myeloblasts, where it mediates effects through antibody-dependent cell-mediated cytotoxicity (ADCC). Cytarabine is a nucleoside analog small molecule chemotherapeutic that inhibits DNA synthesis in proliferating cells. The combination aimed to improve survival by targeting leukemic cells via distinct mechanisms: direct cytotoxicity from cytarabine and immunological targeting from lintuzumab. In randomized, phase IIb trials, the regimen did not improve overall survival over cytarabine alone in elderly patients[1][2][3][5].
