A combination of **lintuzumab**, a humanized monoclonal antibody targeting CD33, and **lenalidomide**, an oral immunomodulatory small molecule. Lintuzumab binds to CD33, a sialic acid–binding immunoglobulin-like lectin expressed on myeloid leukemic blasts and has antileukemic effects via antibody-dependent cell-mediated cytotoxicity. Lenalidomide modulates the immune microenvironment, inhibits angiogenesis, and has direct antineoplastic effects; it is approved for multiple myeloma and certain myelodysplastic syndromes. The combination is under clinical investigation, particularly for myelodysplastic syndromes (MDS), with the hypothesis that simultaneous myeloid blast targeting and immune stimulation may enhance antitumor efficacy.
