Overview

Linvoseltamab + carfilzomib is a combination therapy investigated for treatment of relapsed/refractory multiple myeloma, particularly in patients with prior exposure to multiple drug classes. Linvoseltamab (REGN5458, marketed as Lynozyfic) is a BCMA × CD3 bispecific monoclonal antibody that redirects cytotoxic T cells to multiple myeloma cells expressing B-cell maturation antigen (BCMA), thereby promoting T-cell–mediated tumor cell lysis. Carfilzomib (brand name Kyprolis, code name PR-171) is a second-generation, irreversible proteasome inhibitor that selectively inhibits the chymotrypsin-like activity of the 20S proteasome, leading to apoptosis and growth inhibition of malignant plasma cells. The combination aims for synergistic cytoreduction by enhancing both immunostimulatory and cytotoxic mechanisms, with early clinical data indicating robust response rates and manageable toxicity profiles in heavily pretreated populations[1][4][2][3][5].
