Lipoplatin + verteporfin is a combination therapy regimen investigated by the Centre Hospitalier Universitaire Vaudois (CHUV) for the treatment of malignant pleural effusion (MPE). The treatment consists of Lipoplatin, a liposomal nanoparticle formulation of the platinum-based alkylating agent cisplatin, administered intravenously in conjunction with Visudyne (verteporfin), a benzoporphyrin-derived photosensitizer. The therapeutic approach utilizes low-dose intrapleural photodynamic therapy (PDT), also termed photo-induction, to modulate the tumor microvasculature and enhance the localized uptake of systemically administered Lipoplatin into pleural malignancies. Cisplatin exerts its cytotoxic effect by forming intra-strand and inter-strand DNA crosslinks, which inhibit DNA replication and transcription, leading to cell cycle arrest and apoptosis. Verteporfin-mediated PDT is intended to overcome chemo-resistance and improve local tumor control in patients undergoing video-assisted thoracoscopic surgery (VATS) pleurodesis.
