Liposomal amphotericin B + miltefosine is a combination therapy used primarily for the treatment of visceral leishmaniasis, including in patients co-infected with HIV. Liposomal amphotericin B is a lipid-based formulation of the polyene antifungal and antiprotozoal agent amphotericin B, designed to improve drug delivery to macrophages in affected organs and reduce toxicity compared to conventional formulations. Miltefosine is an oral alkylphosphocholine compound with antileishmanial activity. The combination leverages the rapid parasite clearance of liposomal amphotericin B and the sustained activity of miltefosine, showing synergistic effects in vitro and improved clinical outcomes in some studies. This regimen has been evaluated in phase III trials for visceral leishmaniasis/HIV coinfection[1][2][3][7].
