This is a combination therapy consisting of liposomal amphotericin B (L-AmB), a polyene antifungal and antileishmanial agent formulated in liposomes for improved safety and efficacy, and sodium stibogluconate (SSG), a pentavalent antimonial compound. The combination is primarily used for the treatment of visceral leishmaniasis (VL), especially in regions where monotherapy has limitations due to toxicity, resistance, or cost. Liposomal amphotericin B acts by binding to ergosterol-like molecules in the Leishmania cell membrane, causing increased membrane permeability and cell death. Sodium stibogluconate interferes with parasite metabolism through inhibition of glycolytic enzymes and other mechanisms not fully elucidated. Combination regimens aim to improve cure rates while reducing duration of therapy and minimizing toxicity compared to monotherapies[1][2][3][4]. This regimen has been studied particularly in East Africa as an alternative to longer or more toxic treatments.
