Liposomal amphotericin B + voriconazole is a combination antifungal regimen frequently utilized as a standard of care for the treatment of suspected or confirmed invasive mould infections, such as invasive aspergillosis. The regimen typically involves induction therapy with liposomal amphotericin B, a polyene antifungal that binds to ergosterol in the fungal cell membrane to create pores and cause cell death, followed by consolidation therapy with voriconazole, a triazole that inhibits the fungal enzyme 14-alpha-lanosterol demethylase to block ergosterol synthesis. This sequential or concurrent approach is often used as a comparator in clinical trials for novel antifungal agents, such as Elion Therapeutics' turletricin (EL219), to evaluate efficacy and safety against established therapeutic protocols.
