Drug intelligence / Profile preview
Liposomal bupivacaine is an amide-type local anesthetic encapsulated within multivesicular liposomes (DepoFoam technology) designed for prolonged postsurgical analgesia. Unlike standard bupivacaine hydrochloride, which has a relatively short duration of action, the liposomal formulation allows for a slow, controlled release of the drug, extending pain relief for up to 72 hours. Its mechanism of action involves the reversible blockade of voltage-gated sodium channels on nerve membranes, which inhibits the ionic flux required for the initiation and conduction of nerve impulses. Developed by Pacira BioSciences, it is FDA-approved for various applications including local infiltration, interscalene brachial plexus nerve block, and sciatic nerve block to manage postoperative pain, thereby potentially reducing the need for systemic opioids.
